چکیده

In continuation of our investigation of liver diphosphopyridine nucleotide-linked aldehyde dehydrogenase (1)) we have undertaken a study of various classes of inhibitors of the enzyme. Interest in aldehyde oxidation has been increasing in recent years from several points of view. The classical problem of ethanol oxidation in viva still has many unresolved facets (Z), and the enzymes responsible for the metabolism of aldehydes arising from various amines by the action of monamine oxidase are not well characterized (3). There were several sources of information in the search for inhibitors of aldehyde dehydrogenase. A consideration of structural analogues of either of the two substrates, aldehyde or DPN, was a logical starting point; compounds known to bring about reactions to ethanol in man and animals proved to be a rich source of inhibitors; and compounds known to inhibit other aldehyde-oxidizing enzymes were of interest.

Aldehyde dehydrogenase, a diphosphopyridine nucleotide-linked enzyme.

The dismutation of acetaldehyde to ethyl alcohol and acetic acid has been shown to occur in animal tissues (1, 2). The dependence of this reaction on diphosphopyridine nucleotide was demonstrated by von Euler and Brunius (3). Partial purification of the responsible enzyme system was achieved by Dixon and Lutwak-Mann (4). These authors clearly demonstrated that the mutase activity was independen...

A study of the mechanism by which steroid hormones influence rabbit liver aldehyde dehydrogenase.

It has recently been reported (1) that rabbit liver contains a diphosphopyridine nucleotide-linked aldehyde dehydrogenase that is sensitive to a variety of steroid hormones and to the estrogen analogue diethylstilbestrol. One group of steroids uniformly inhibited the reaction; another group of steroids and diethylstilbestrol either inhibited or stimulated, depending upon the experimental condit...

Metabolism of the retina. III. The role of reduced triphoshopyridine nucleotide in the visual cycle.

Exposure of a dark-adapted retina to light results in the liberation of vitamin A aldehyde (retinene) from the visual pigment, rhodopsin, and in subsequent enzymatic reduction of the liberated aldehyde to vitamin A (1). The enzyme, alcohol dehydrogenase (retinene reductase), is present in visual cell outer segments and can use reduced diphosphopyridine nucleotide (DPNH) for the reduction of vit...

Automated assay of activities of enzymes involving the diphosphopyridine nucleotide-reduced diphosphopyridine nucleotide reaction.

A continuous flow instrumental system of analysis, first described by Skeggs (l), was previously applied to the determination of phosphohexose isomerase activity and resulted in a method that was approximately eightfold as rapid as the manual procedure (2). The present paper is concerned with the application of this system of analysis to the determination of enzyme activities that utilize the d...

Enzymatic oxidation of vitamin A aldehyde to vitamin A acid.

Studies indicating that vitamin A (1) and vitamin A aldehyde (2) are metabolized to water-soluble degradation products and CO% provide support for the suggestion (3) that there exists an oxidative pathway of major importance in the metabolism of vitamin A. The detection of vitamin A aldehyde in the liver (4, 5) and observations (6-8) that dietary vitamin A acid supports the growth of vitamin A-...
